Peter Parker on koulukiusattu silmälasipäinen lukiolaisnörtti, joka on ihastunut kauniiseen Mary Janeen. Kun Peterin luokka on luokkaretkellä tutkimuslaitoksessa, häntä puraisee geenimanipuloitu hämähäkki. Pian Peter huomaa että hänellä on omituisia kykyjä, muun muassa hämähäkin nopeus, voima ja ketteryys.
