Kun valtakunnan etsityin ja hurmaavin roisto Flynn Rider piiloutuu arvoitukselliseen torniin, hänet ottaa panttivangiksi Tähkäpää, kaunis ja kipakka torniin vangittu teini, jolla on 20 metriä pitkät, taianomaiset ja kullanvaaleat hiukset. Flynnin utelias vangitsija, joka etsii keinoa päästä pois tornista, jossa hän on ollut vuosia lukkojen takana, tekee sopimuksen komean varkaan kanssa. Niin tämä odottamaton kaksikko lähtee toiminnantäyteiseen seikkailuun, jota täydentävät superpoliisihevonen Maximus, ylisuojeleva kameleontti Pascal ja joukko karkeita tavernaroistoja.

Liukaskielinen Tittelintuure ylipuhuu vanhoja loistonsa päiviä kaihoavan Shrekin allekirjoittamaan houkuttelevan sopimuksen. Pian Shrek löytääkin itsensä vääristyneestä Kaukaisen maan käänteisversiosta, jossa jättejä jahdataan, Tittelintuure on kuningas eivätkä Shrek ja Fiona ole koskaan tavanneet. Shrekin tehtävänä on nyt saada tehty tekemättömäksi pelastaakseen ystävänsä, saadakseen vanhan elämänsä takaisin ja saavuttaakseen taas Fionan rakkauden.

Boo Boo on Jogin uskollinen ystävä ja apuri Jogin lukemattomissa juonissa, kun karhukaksikko havittelee käpäliinsä Jellystonen vierailijoiden lounaita. Jellystonen kansallispuiston kävijämäärä on laskenut, joten pormestari Brown on päättänyt sulkea puiston ja myydä sen maat. Se tarkoittaisi, että ihmiset eivät voisi enää nauttia ulkoilmaelämästä ja luonnon kauneudesta − ja mikä pahinta, Jogi ja hänen kumppaninsa Boo Boo potkittaisiin ulos ainoasta kodista, joka heillä on koskaan ollut. Elämänsä suurimman haasteen kohtaavan Jogin on todistettava, että hän todella on ”fiksumpi kuin keskivertokarhu”, kun hän ja Boo Boo yhdistävät voimansa metsänvartija Smithin kanssa löytääkseen keinon estää puiston sulkeminen ikiajoiksi.
