Sean (Kerr Smith) er blitt kastet ut i en sjebnesvanger kamp mot tiden. En kamp som kommer til å avgjøre hans fremtid fo alltid, da han blir infisert av et virus, som til slutt vil forvandle ham til en blodsugende, kjøttetende vampyr. Under en kjøretur tvers over USA tar sjåføren Sean en farlig omvei. Han tar med Nick (Brendan Fehr), en haiker, som er på sporet av en gruppe vampyrer. Ved en motorveikafeterisa møter de en smårar og hjelpesløs kvinne, som nettopp har vært utsatt for vampyrenes blodrus, og Sean blir smittet. Nå blir de alle tre kastet ut i en desperat kamp mot tiden for å oppspore og drepe sykdommens kilde: århundrets vampyr med overmenneskelig styrke og en umettelig tørst etter blod.
