Iso-Britanniassa kolme eläinaktivistia murtautuu laboratorioon vapauttaakseen siellä olevat sairaat simpanssit. He kohtaavat laborantin, joka kertoo simpanssien kantavan virusta nimeltä "raivo". Aktivistit tuhahtavat eivätkä ota todesta laborantin varoituksia, ja vapauttavat yhden simpansseista. 28 päivää myöhemmin pyörälähetti nimeltä Jim herää tyhjästä sairaalasta Lontoossa ja alkaa ihmetellä, missä on kaikki ihmiset ja miksi koko kaupunki on tyhjä?
